Mutants' Stabilities

Ala16 and Leu49 are two of the most important residues constituting the main hydrophobic core of WT-CI2.A. GdnHCl denatured
fluorescence curve.

Left: LA49(I)+WT(II)
Right: AG16(I)+WT(II)
(Ex 280nm, Em 356nm)

B.By stopped-flow fluorescence:

(Left hand side)
Top:
LA49(I)+WT(II) and AG16(I)+WT(II)
Meddle: "Slower phase" of
LA49(I)+WT(II)
Bottom: "Slower phase" of
AG16(I)+WT(II)
The result showed that LA49(I)+WT(II) and AG16(I)+WT(II) were described by two-state transition, and they did not populate a stable intermediate.
