Different cleavage sites are found in SNAREs as the targets of clostridial neurotoxin. All neurotoxin-mediated cleavage sites are located between the C-terminal membrane anchors and the ionic “0” layer.
Only uncomplexed or partially assembled SNARE proteins can undergo proteolysis because the protease-cleavage sites or protease-recognition sites are protected upon complex formation as mention previously.
The cleavage may inhibit the formation of complex and the membrane fusion between vesicle and synapse resulting the blockage of neurotransmitter transportation.
